Ipamorelin – what the peptide is

Ipamorelin is a synthetic pentapeptide from the group of growth hormone-releasing peptides. Unlike GHRH analogs, it acts on the ghrelin receptor, also known as the growth hormone secretagogue receptor.

The molecule is described as selective – the basis for this designation is that, within the studied ranges, it triggers release of growth hormone without pronounced effect on cortisol and prolactin, unlike earlier compounds in the group.

This selectivity is also the reason the peptide is considered separately from GHRP-6 and GHRP-2, which originate from the same family but have a broader hormonal profile.

Mechanism of action

Binding to the ghrelin receptor in the pituitary activates a signaling pathway distinct from that of the GHRH receptor. The two pathways act synergistically, which is the basis for research into their combined activation.

The ghrelin receptor is also found in the hypothalamus, where it participates in appetite regulation. With Ipamorelin, however, the appetite component is described as weakly expressed – a difference compared to GHRP-6, in which it is more pronounced.

The selectivity is attributed to the structure of the molecule, which limits interaction with receptor systems associated with cortisol release.

Research data and observations

Observations track growth hormone concentrations following administration and compare them with cortisol and prolactin values. This comparison is the basis of the selectivity designation.

A separate line of research examines combination with a GHRH analog. Since the two compounds act through different receptors, the discussion focuses on whether the response exceeds the sum of the individual effects.

Much of the data originates from preclinical models and from studies with a limited number of participants.

When interpreting published data, the distinction between preclinical and clinical conditions should be taken into account. Results from cell cultures and animal models describe a mechanism but do not allow quantitative conclusions for human conditions.

The dose ranges used in preclinical studies are often not directly translatable due to differences in metabolism between species. This limitation is consistently noted in review papers.

Example calculations

The calculations below are purely mathematical examples showing the relationship between total substance amount, solvent volume, and resulting concentration. They do not represent a dosing recommendation, administration schedule, or instructions for use.

When dissolving 10 mg with 2 ml bacteriostatic water:

The resulting concentration is 5 mg/ml, with 0.20 ml containing 1 mg of substance.

Review of scientific publications

The literature on selective ghrelin agonists is smaller in volume than that on earlier representatives of the group, since these compounds were described later.

Comparison between individual secretagogues requires care due to differences in receptor selectivity and in the studied ranges.

Storage and stability

Our peptides are manufactured in compliance with strict quality standards and controls. Each product is supplied in lyophilized form, which ensures stability and prevents quality degradation during transport. We test every batch.

The independent analysis protocol from Janoshik Analytical for the specific batch is published directly beneath the main product photo. The document contains the result of the identity and purity testing of the substance and can be reviewed before ordering.

Lyophilized peptides are stored in the original vial, protected from light and moisture. In this state, their stability is considerably higher than after reconstitution, since the absence of water slows the processes of hydrolysis and aggregation of the peptide chain.

After reconstitution with bacteriostatic water, the solution is stored refrigerated. Bacteriostatic water contains a preservative that limits microbial growth, unlike sterile water for injections. Repeated freezing and thawing is described in the literature as a factor that accelerates degradation of the peptide chain and leads to loss of initial purity.

Safety data from studies

Reported observations include reactions at the injection site and a brief sensation of warmth. The profile is described as more limited compared to earlier compounds in the group.

Long-term controlled studies in humans have not been published.

A general limitation for compounds in this category is the absence of long-term controlled studies in humans. Available information describes mechanism and short-term observations, not a safety profile for prolonged use.
