IGF-1 LR3 – what the peptide is

IGF-1 LR3 is a modified analog of insulin-like growth factor-1 – a natural 70-amino-acid peptide that mediates much of the growth hormone's action on peripheral tissues.

The designation LR3 describes two modifications: an extension with an additional 13 amino acids at the beginning of the chain and a substitution at position 3. The purpose of these changes is to reduce binding to the binding proteins that, under natural conditions, retain most of the circulating IGF-1.

Since the binding proteins determine how much of the hormone is available to the receptors, their reduced affinity for the analog substantially changes its pharmacokinetic profile compared to the natural molecule.

Mechanism of action

The molecule binds to the IGF-1 receptor – a tyrosine kinase receptor structurally related to the insulin receptor. Its activation triggers intracellular signaling pathways involved in cell growth and protein synthesis.

The reduced binding to the binding proteins means that a larger proportion of the molecule remains in free form. This is also the main described difference compared to natural IGF-1.

Due to the structural similarity between the IGF-1 and insulin receptors, cross-binding is also described, which underlies the observations regarding glucose metabolism.

Research data and observations

Observations examine the effect on protein synthesis and cell proliferation in preclinical models. It is noted that the response depends on the tissue type and receptor availability.

A separate line of research tracks the effect on blood glucose resulting from cross-binding with the insulin receptor.

Much of the data originates from cell cultures and animal models.

When interpreting published data, the difference between preclinical and clinical conditions should be taken into account. Results from cell cultures and animal models describe mechanism but do not allow quantitative conclusions regarding human conditions.

The dose ranges used in preclinical studies are often not directly translatable due to differences in metabolism between species. This limitation is consistently noted in review papers.

Example calculations

The calculations below are purely mathematical examples illustrating the relationship between the total amount of substance, the volume of solvent and the resulting concentration. They do not constitute a dosing recommendation, administration schedule or instructions for use.

Overview of scientific publications

The literature on natural IGF-1 is extensive, while that on modified analogs is more limited and focused mainly on biotechnological applications in cell cultures.

Results pertaining to the natural molecule are not directly transferable to the analog due to differences in binding.

Safety data from studies

The main described consideration is the effect on blood glucose resulting from binding to the insulin receptor. A concentration-dependent relationship is noted.

Systematized data on tolerability in humans have not been published in the available literature.

A general limitation for compounds in this category is the absence of long-term controlled studies in humans. The available information describes mechanism and short-term observations, not a safety profile for prolonged use.
