IGF-1 LR3 – what the peptide is

IGF-1 LR3 is a modified analog of insulin-like growth factor-1 – a natural 70-amino-acid peptide that mediates much of the action of growth hormone on peripheral tissues.

The designation LR3 describes two modifications: an extension with 13 additional amino acids at the beginning of the chain and a substitution at position 3. The purpose of these changes is to reduce binding to the binding proteins that, under natural conditions, sequester most of the circulating IGF-1.

Since the binding proteins determine how much of the hormone is available to the receptors, the reduced affinity of the analog for them substantially alters its pharmacokinetic profile compared to the natural molecule.

Mechanism of action

The molecule binds to the IGF-1 receptor – a tyrosine kinase receptor structurally close to the insulin receptor. Its activation triggers intracellular signaling pathways involved in cell growth and protein synthesis.

The reduced binding to binding proteins means that a greater proportion of the molecule remains in free form. This is also the main described difference compared to natural IGF-1.

Due to the structural similarity between the IGF-1 and insulin receptors, cross-binding is also described, which underlies the observations regarding glucose metabolism.

Research data and observations

Observations examine the influence on protein synthesis and cell proliferation in preclinical models. It is noted that the response depends on the tissue type and receptor availability.

A separate line of research follows the effect on blood glucose resulting from cross-binding with the insulin receptor.

Most of the data originate from cell cultures and animal models.

When interpreting the published data, the difference between preclinical and clinical conditions should be taken into account. Results from cell cultures and animal models describe mechanism but do not allow quantitative conclusions for human conditions.

The dosage ranges used in preclinical studies are often not directly translatable due to differences in metabolism between species. This limitation is consistently noted in review papers.

Example calculations

The calculations below are purely mathematical examples showing the relationship between the total amount of substance, the volume of solvent, and the resulting concentration. They do not represent a recommendation for dosing, an administration schedule, or instructions for use.

The resulting concentration is 1 mg/ml, with 1.00 ml containing 1 mg of substance.

Overview of scientific publications

The literature on natural IGF-1 is extensive, while that on modified analogs is more limited and focuses mainly on biotechnological applications in cell cultures.

Results relating to the natural molecule do not directly transfer to the analog due to differences in binding.

Storage and stability

Our peptides are manufactured in compliance with strict quality standards and control. Each product is supplied in lyophilized form, which ensures stability and prevents quality degradation during transport. We test every batch.

The independent analysis report from Janoshik Analytical for the specific batch is published directly below the main product photo. The document contains the results of the identity and purity testing of the substance and can be reviewed before ordering.

Lyophilized peptides should be stored in the original vial, protected from light and moisture. In this state their stability is significantly higher than after reconstitution, since the absence of water slows down the processes of hydrolysis and aggregation of the peptide chain.

After reconstitution with bacteriostatic water, the solution should be stored refrigerated. Bacteriostatic water contains a preservative that limits microbial growth, unlike sterile water for injection. Repeated freezing and thawing is described in the literature as a factor that accelerates degradation of the peptide chain and leads to loss of the original purity.

Safety data from studies

The main consideration described is the effect on blood glucose resulting from binding to the insulin receptor. A concentration dependence is noted.

Systematic data on tolerability in humans have not been published in the available literature.

A general limitation for compounds in this category is the absence of long-term controlled studies in humans. The available information describes mechanism and short-term observations, not a safety profile for prolonged use.
