CJC-1295 – what the peptide is

CJC-1295 is a modified analog of growth hormone-releasing hormone, built on the GHRH (1-29) sequence. The compound carries four amino acid substitutions, aimed at protecting it from the rapid degradation to which the natural molecule is subjected.

The main target of this degradation is the enzyme dipeptidyl peptidase-4, which cleaves the initial amino acids of GHRH within minutes. The substitutions in CJC-1295 slow down this process and extend the molecule's presence in circulation.

The no-DAC variant does not carry the additional linking group that, in the other form, allows attachment to albumin. This is also why the two forms are considered separately in the literature – they differ substantially in duration of action.

Mechanism of action

The molecule binds to the GHRH receptor in the anterior pituitary and triggers release of stored growth hormone. The mechanism is identical to that of natural GHRH, with the difference lying in the time profile.

Since regulation remains subject to somatostatin, release retains a pulsatile character. In the literature this is described as a difference compared to the continuous presence seen with direct administration of growth hormone.

The no-DAC form has a significantly shorter action than the DAC form, which explains why the two are studied as separate compounds rather than interchangeable variants.

Research data and observations

Observations track growth hormone and IGF-1 concentrations following administration. The no-DAC form is noted to produce more pronounced but shorter-lasting changes compared to the long-acting form.

A separate line of work examines the combination of a GHRH analog with a ghrelin receptor agonist. The two groups act through different receptors, and discussion centers on whether their effects are additive.

Individual variability in response is significant and is attributed to differences in pituitary reserve.

When interpreting published data, the difference between preclinical and clinical conditions should be taken into account. Results from cell cultures and animal models describe mechanism but do not allow quantitative conclusions for human conditions.

The dose ranges used in preclinical studies are often not directly translatable due to differences in metabolism between species. This limitation is consistently noted in review papers.

Sample calculations

The calculations below are purely mathematical examples showing the relationship between total substance amount, solvent volume, and resulting concentration. They do not represent a dosing recommendation, administration schedule, or instructions for use.

Overview of scientific publications

The literature on modified GHRH analogs is more limited than that on the natural hormone. Some publications do not clearly distinguish between the two forms of CJC-1295, which complicates comparison.

Review papers recommend explicitly specifying the studied form when citing results.

Storage and stability

Our peptides are manufactured in compliance with strict quality control standards. Each product is supplied in lyophilized form, which ensures stability and prevents quality degradation during transport. We test every batch.

The independent analysis report from Janoshik Analytical for the specific batch is published directly below the main product photo. The document contains the results of identity and purity testing of the substance and can be reviewed before ordering.

Lyophilized peptides are stored in the original vial, protected from light and moisture. In this state their stability is significantly higher than after reconstitution, since the absence of water slows down hydrolysis and aggregation processes of the peptide chain.

After reconstitution with bacteriostatic water, the solution is stored refrigerated. Bacteriostatic water contains a preservative that limits microbial growth, unlike sterile water for injection. Repeated freezing and thawing is described in the literature as a factor that accelerates degradation of the peptide chain and leads to loss of original purity.

Safety data from studies

Reported observations include reactions at the administration site and short-term manifestations related to vasodilation. A concentration-dependent relationship has been noted.

Systematized data from long-term studies in humans have not been published in the available literature.

A general limitation for compounds in this category is the absence of long-term controlled studies in humans. Available information describes mechanism and short-term observations, not a safety profile for prolonged use.
